Lanosidade em cultivares de pêssego (Prunus persica (L.) Batsch): estudos anatômicos e ultra-estruturais Rev. Bras. Bot.

A fim de detectar o período de manifestação dos sintomas de lanosidade e compreender melhor as alterações celulares que acompanham a injúria causada pelo frio, foram analisados dois cultivares de pêssegos: 'Aurora-1' (não sensível ao frio) e 'Dourado-2' (sensível ao frio), durante o armazenamento refrigerado a 3 ºC seguido da comercialização simulada por dois dias a 25 ºC. Foram feitas análises aos microscópios fotônico e eletrônico de transmissão. O mesocarpo dos pêssegos, no momento da colheita, foi anatomicamente similar e apresentou células parenquimáticas túrgidas isodiamétricas de tamanhos variáveis arranjadas com amplos espaços intercelulares. In the Brazilian Cerrado (neotropical savanna), the development of bud-bearing underground systems as adaptive structures to fire and dry periods can comprise an important source of buds for this ecosystem, as already demonstrated in the Brazilian Campos grasslands and North American prairies. Asteraceae species from both woody and herbaceous strata have subterranean organs that accumulate carbohydrates, reinforcing the adaptive strategy of these plants to different environmental conditions. Ultrastructural analysis of in vitro direct and indirect organogenesis Rev. Bras. Bot.

This is a comparative study of the ultrastructural characteristics of the cells involved in the organogenesis in vitro of Bauhinia forficata Link (indirect system) and Glycine max (L.) Merrill (direct system). B. forficata calli after 30 days culture and G. max meristemoids after 10 days culture were prepared for ultrastructural analysis using conventional methods. Concentrically arranged rough endoplasmic reticulum (RER) and plastids containing starch grains were seen during G. max and B. forficata organogenesis. The amitotic process, the presence of plastids around the nucleus and nuclear envelope with conspicuous pores were found in B. forficata.

O presente trabalho objetivou avaliar as características anatômicas e físico-químicas de pêssegos (Prunus persica (L.) Batsch) 'Aurora-1' e 'Dourado-2', armazenados em diferentes temperaturas e períodos. No primeiro experimento, os frutos foram armazenados a 0, 3 e 6ºC por 14, 21, 28 e 35 dias (mais dois dias de simulação à comercialização, sob 25ºC). Anatomical confirmation of root parasitism in Brazilian Agalinis Raf. species (Scrophulariaceae) BABT

Agalinis Raf. consists approximately of 60 species, 14 of which occur in Brazil. The genus presents predominantly american distribution and the Brazilian species appears mainly in high areas of Minas Gerais. The North-American species are refered as hemiparasites, but there is no anatomical data about it in relation to the Brazilian species. Anatomical studies were conducted to verify whether the Agalinis species from Brazil were root parasites or not. Anatomy of somatic embryogenesis in Carica papaya L. BABT

Mature zygotic embryos of Carica papaya L. ‘Sunrise Solo’ were used as explants for embryogenesis induction. The explants were inoculated on Murashige and Skoog culture medium supplemented with 2 mg.L-1 2,4-dichlorophenoxyacetic acid and incubated in darkness at 25+2°C. Histological analysis of callogenesis and somatic embryogenesis indicated occurrence of direct and indirect somatic embryogenesis development. The sugarcane spittlebug Mahanarva fimbriolata (Stål) (Hemiptera: Cercopidae) is a pest of mechanically-harvested sugarcane in Brazil, when trash burning is not performed. To better understand the differences in feeding behavior of adults and nymphs of this pest and the subsequent disorders that arise, stylet penetration through fixation, staining and sectioning was investigated. Nymphs cause a "physiological disorder" damaging the tracheary system of the roots, slowing or preventing water and nutrient flow, with phloem and xylem dehydration. This work dealt with the anatomy of the underground system in Vernonia grandiflora Less. and V. brevifolia Less. (Vernonieae; Asteraceae), two perennial geophytes, to elucidate their ability to sprout in the Brazilian Cerrado conditions. V. grandiflora, a subshrubby species, possessed a thickened underground system constituted by a xylopodium and many tuberous roots. Different concentrations of 6-benzyladenine (6-BA) were used in vitro to induce buds in three types of explants: leaf discs, root segments and the seedling itself obtained from in vitro seed germination of Passiflora cincinnata Mast. O objetivo deste trabalho foi apresentar a descrição anatômica dos tecidos lesionados pelo Citrus leprosis virus do tipo citoplasmático (CiLV-C), em ramos e folhas de laranjeira-doce (Citrus sinensis (L.) Osbeck) 'Pêra', e fornecer dados para a compreensão das interações entre o vírus e a planta hospedeira. Tecidos que apresentavam lesões foram seccionados em micrótomo rotativo (5-7 µm de espessura) e, posteriormente, as lâminas foram coradas e montadas em resina sintética. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological analysis of the callogenesis and organogenesis from root segments of Curcuma zedoaria Roscoe BABT

Callus was induced from root segments taken from in vitro grown plants of Curcuma zedoaria Roscoe. The explants were cultured on agar-solidified Murashige and Skoog medium supplemented with 13.4muM of alpha-naphthaleneacetic acid and 2.2muM of 6-benzylaminopurine at 25ºC in the dark. Histological analysis revealed that callus was formed from the hypertrophied cortical parenchyma cells of the explant. Some of these cells underwent division while the surrounding cells accumulated starch. Comparative leaf morphology and anatomy of three Asteraceae species BABT

The objective of this paper was to describe and compare the morphology and anatomy of mature leaves of Mikania glomerata Spreng., Porophyllum ruderale Cass. and Vernonia condensata Baker (Asteraceae) species that have different habits emphasizing their secretory structures. Longitudinal and transversal sections of mature leaf blades of the three species were analyzed at the apex, base, and medium third part of the midvein of the leaf blade and of the margin. M. glomerata had uniseriate glandular trichomes and secretory ducts; P. ruderale had hydathodes and secretory cavities; and V. condensata had idioblasts and uni-and biseriate glandular trichomes.

Plant regeneration from proroplasts of alfalfa (Medicago sativa) via somatic embryogenesis Scientia Agricola

Alfalfa is one of the most frequently studied species from the production of tissue culture-derived embryos point of view. In this study, five alfalfa cultivars were analyzed with reference to their ability to regenerate plants from protoplast cultures via somatic embryogenesis. Plant regeneration from leaf-derived protoplasts isolated from the cultivar Rangelander was achieved using a protocol defined for alfalfa cell suspension-derived embryogenesis. Because of its high efficiency, this procedure is recommended for protoplast electroporation-mediated genetic transformation of alfalfa.

Gomphrena macrocephala St.-Hil. (Amaranthaceae) is a perennial herb from the cerrado with medicinal properties and ornamental interest. Plants can be micropropagated through nodal segments, but acclimatization is difficult. The aim of this study was to establish a relationship between anatomical aspects of the roots and the acclimatization process. When cultures were supplemented with IBA, callus and thick and frangible roots appeared at the base of the microcuttings. A fragile vascular connection between roots and shoots was observed.
